Technical note
Dieses ELISA-Kit verwendet das kompetitive ELISA-Prinzip. Die in diesem Kit enthaltene Mikro-ELISA-Platte ist mit humanem T vorbeschichtet. Proben (oder Standards) und Meerrettichperoxidase (HRP)-gekoppelte Antikörper, die spezifisch für humanes T sind, werden in die Vertiefungen der Mikro-ELISA-Platte gegeben. Humanes T in Proben (oder Standards) konkurriert mit einer festen Menge an T auf dem Festphasenträger um Bindungsstellen am HRP-gekoppelten Detektionsantikörper, der spezifisch für T ist. Überschüssiges Konjugat und ungebundene Probe oder Standard werden von der Platte gewaschen. Die Substratlösung wird zu jeder Vertiefung gegeben. Die Enzym-Substrat-Reaktion wird durch Zugabe der Stopplösung beendet und die Farbe wird gelb. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450±2 nm gemessen. Die Konzentration von humanem T in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
