Technical note
Dieser ELISA-Kit nutzt das Competitive-ELISA-Prinzip. Die in diesem Kit enthaltene Mikro-ELISA-Platte ist mit humanem Cortisol vorbeschichtet. Proben (oder Standards) und ein Meerrettichperoxidase (HRP)-gekoppelte Antikörper, der spezifisch für humanes Cortisol ist, werden den Vertiefungen der Mikro-ELISA-Platte hinzugefügt. Humanes Cortisol in den Proben (oder Standards) konkurriert mit einer festen Menge an Cortisol auf dem festphasengebundenen Träger um Bindungsstellen am HRP-gekoppelten Detektionsantikörper, der spezifisch für Cortisol ist. Überschüssiges Konjugat und ungebundene Probe oder Standard werden von der Platte gewaschen. Die Substratlösung wird zu jeder Vertiefung hinzugefügt. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Stopplösung beendet, woraufhin sich die Farbe gelb färbt. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450 ± 2 nm gemessen. Die Konzentration des humanen Cortisols in den Proben wird anschließend durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
