Technical note
Dieses ELISA-Kit verwendet das kompetitive ELISA-Prinzip. Die in diesem Kit enthaltene Mikro-ELISA-Platte wurde mit Hühner-E3 vorbeschichtet. Proben (oder Standards) und Meerrettichperoxidase (HRP)-gekoppelte Antikörper, die spezifisch für Hühner-E3 sind, werden in die Vertiefungen der Mikro-ELISA-Platte gegeben. Hühner-E3 in Proben (oder Standards) konkurriert mit einer festgelegten Menge an E3 auf dem festen Träger um Bindungsstellen am HRP-gekoppelten Detektionsantikörper, der spezifisch für E3 ist. Überschüssiges Konjugat und ungebundene Probe oder Standard werden von der Platte gewaschen. Die Substratlösung wird zu jeder Vertiefung hinzugefügt. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Stopplösung beendet und die Farbe wird gelb. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450±2 nm gemessen. Die Konzentration von Hühner-E3 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
