Technical note
Dieser ELISA-Kit nutzt das Prinzip des kompetitiven ELISA. Die in diesem Kit enthaltene Mikro-ELISA-Platte wurde mit Hunde-Pg vorbeschichtet. Proben (oder Standards) und Meerrettichperoxidase (HRP)-gekoppelte Antikörper, die spezifisch für Hunde-Pg sind, werden zu den Mikro-ELISA-Plattenvertiefungen gegeben. Hunde-Pg in Proben (oder Standards) konkurriert mit einer festen Menge an Pg auf dem Festphasenträger um Bindungsstellen am HRP-gekoppelten Detektionsantikörper, der spezifisch für Pg ist. Überschüssiges Konjugat und ungebundene Proben oder Standards werden von der Platte gewaschen. Die Substratlösung wird zu jeder Vertiefung hinzugefügt. Die Enzym-Substrat-Reaktion wird durch Zugabe der Stopplösung beendet und die Farbe wird gelb. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450±2 nm gemessen. Die Konzentration von Hunde-Pg in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
