Technical note
Dieses ELISA-Kit nutzt das Sandwich-ELISA-Prinzip. Die in diesem Kit enthaltene Mikro-ELISA-Platte ist mit einem Antikörper vorbeschichtet, der spezifisch für Maus-TFPI ist. Standards oder Proben werden zu den Vertiefungen der Mikro-ELISA-Platte hinzugefügt und verbinden sich mit dem spezifischen Antikörper. Anschließend werden nacheinander ein biotinylierter Detektionsantikörper, der spezifisch für Maus-TFPI ist, und ein Avidin-Meerrettichperoxidase (HRP)-Konjugat zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nicht gebundene Komponenten werden abgewaschen. Die Substratlösung wird zu jeder Vertiefung gegeben. Nur jene Vertiefungen, die Maus-TFPI, biotinylierten Detektionsantikörper und Avidin-HRP-Konjugat enthalten, erscheinen blau. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Stopplösung beendet, woraufhin sich die Farbe gelb färbt. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 13 nm gemessen. Der OD-Wert ist proportional zur Konzentration von Maus-TFPI. Die Konzentration von Maus-TFPI in den Proben kann durch Vergleich der OD der Proben mit der Standardkurve berechnet werden.
