Technical note
Dieses ELISA-Kit basiert auf dem Sandwich-ELISA-Prinzip. Die in diesem Kit enthaltene Mikro-ELISA-Platte ist mit einem CD40-spezifischen Antikörper vorbeschichtet. Standards oder Proben werden in die Vertiefungen der Mikro-ELISA-Platte gegeben und mit dem spezifischen Antikörper kombiniert. Anschließend werden nacheinander ein biotinylierter Nachweisantikörper, der spezifisch für CD40 ist, und ein Avidin-Meerrettichperoxidase (HRP)-Konjugat zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Ungestattete Komponenten werden abgewaschen. Die Substratlösung wird zu jeder Vertiefung hinzugefügt. Nur jene Vertiefungen, die CD40, den biotinylierten Nachweisantikörper und das Avidin-HRP-Konjugat enthalten, erscheinen blau. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Stopplösung beendet, und die Farbe wechselt zu Gelb. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 47 nm gemessen. Der OD-Wert ist proportional zur CD40-Konzentration. Die CD40-Konzentration in den Proben kann durch Vergleich der OD-Werte der Proben mit der Standardkurve berechnet werden.
