Technical note
Dieses ELISA-Kit verwendet das Sandwich-ELISA-Prinzip. Die in diesem Kit enthaltene Mikro-ELISA-Platte wurde mit einem Antikörper vorbeschichtet, der spezifisch für gp130 ist. Standards oder Proben werden zu den Vertiefungen der Mikro-ELISA-Platte hinzugefügt und mit dem spezifischen Antikörper kombiniert. Dann werden nacheinander ein biotinylierter Nachweisantikörper, der spezifisch für gp130 ist, und ein Avidin-Meerrettichperoxidase (HRP)-Konjugat zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Freie Komponenten werden abgewaschen. Die Substratlösung wird zu jeder Vertiefung hinzugefügt. Nur jene Vertiefungen, die gp130, biotinylierten Nachweisantikörper und Avidin-HRP-Konjugat enthalten, erscheinen blau. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Stopplösung beendet, woraufhin die Farbe gelb wird. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 2 nm gemessen. Der OD-Wert ist proportional zur Konzentration von gp130. Sie können die Konzentration von gp130 in den Proben berechnen, indem Sie die OD der Proben mit der Standardkurve vergleichen.
