Technical note
Dieses ELISA-Kit basiert auf dem Sandwich-ELISA-Prinzip. Die in diesem Kit enthaltene Mikro-ELISA-Platte ist mit einem Antikörper vorbeschichtet, der spezifisch für Ratten-FAS/CD95 ist. Standards oder Proben werden zu den Vertiefungen der Mikro-ELISA-Platte hinzugefügt und mit dem spezifischen Antikörper kombiniert. Anschließend werden nacheinander ein biotinylierter Detektionsantikörper, der spezifisch für Ratten-FAS/CD95 ist, und ein Avidin-Meerrettichperoxidase (HRP)-Konjugat zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Freie Komponenten werden abgewaschen. Die Substratlösung wird zu jeder Vertiefung hinzugefügt. Nur jene Vertiefungen, die Ratten-FAS/CD95, den biotinylierten Detektionsantikörper und das Avidin-HRP-Konjugat enthalten, erscheinen blau. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Stopplösung beendet und die Farbe wechselt zu Gelb. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 2 nm gemessen. Der OD-Wert ist proportional zur Konzentration von Ratten-FAS/CD95. Die Konzentration von Ratten-FAS/CD95 in den Proben kann durch Vergleich der OD der Proben mit der Standardkurve berechnet werden.
