Technical note
Dieses ELISA-Kit basiert auf dem Sandwich-ELISA-Prinzip. Die in diesem Kit enthaltene Mikro-ELISA-Platte ist mit einem spezifischen Antikörper für Maus-POSTN/OSF-2 vorbeschichtet. Standards oder Proben werden in die Vertiefungen der Mikro-ELISA-Platte gegeben und mit dem spezifischen Antikörper kombiniert. Anschließend werden nacheinander ein biotinylierter Nachweisantikörper, spezifisch für Maus-POSTN/OSF-2, und ein Avidin-Meerrettichperoxidase (HRP)-Konjugat zu jeder Vertiefung der Mikroplatte gegeben und inkubiert. Ungenutzte Komponenten werden abgewaschen. Die Substratlösung wird jeder Vertiefung zugegeben. Nur jene Vertiefungen, die Maus-POSTN/OSF-2, den biotinylierten Nachweisantikörper und das Avidin-HRP-Konjugat enthalten, erscheinen blau. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Stopplösung beendet, und die Farbe wechselt zu Gelb. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 2 nm gemessen. Der OD-Wert ist proportional zur Konzentration von Maus-POSTN/OSF-2. Die Konzentration von Maus-POSTN/OSF-2 in den Proben kann durch Vergleich der OD der Proben mit der Standardkurve berechnet werden.
