Technical note
Dieser ELISA-Kit nutzt das Sandwich-ELISA-Prinzip. Die in diesem Kit enthaltene Mikro-ELISA-Platte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Maus NOS3/eNOS ist. Standards oder Proben werden zu den Vertiefungen der Mikro-ELISA-Platte gegeben und mit dem spezifischen Antikörper kombiniert. Anschließend werden ein biotinylierter Detektionsantikörper, der spezifisch für Maus NOS3/eNOS ist, und ein Avidin-Meerrettichperoxidase (HRP)-Konjugat nacheinander zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Freie Komponenten werden abgewaschen. Die Substratlösung wird zu jeder Vertiefung gegeben. Nur jene Vertiefungen, die Maus NOS3/eNOS, biotinylierten Detektionsantikörper und Avidin-HRP-Konjugat enthalten, erscheinen blau. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Stopplösung beendet, und die Farbe wird gelb. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 2 nm gemessen. Der OD-Wert ist proportional zur Konzentration von Maus NOS3/eNOS. Die Konzentration von Maus NOS3/eNOS in den Proben kann durch Vergleich der OD der Proben mit der Standardkurve berechnet werden.
