Technical note
Dieser ELISA-Kit nutzt das Sandwich-ELISA-Prinzip. Die in diesem Kit enthaltene Mikro-ELISA-Platte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Maus bFGF/FGF2 ist. Standards oder Proben werden in die Mikro-ELISA-Plattenvertiefungen gegeben und mit dem spezifischen Antikörper kombiniert. Anschließend werden nacheinander ein biotinylierter Nachweisantikörper, der spezifisch für Maus bFGF/FGF2 ist, und ein Avidin-Meerrettichperoxidase (HRP)-Konjugat zu jeder Mikroplattenvertiefung gegeben und inkubiert. Freie Komponenten werden abgewaschen. Die Substratlösung wird zu jeder Vertiefung gegeben. Nur jene Vertiefungen, die Maus bFGF/FGF2, den biotinylierten Nachweisantikörper und das Avidin-HRP-Konjugat enthalten, erscheinen blau. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Stopplösung beendet und die Farbe wechselt zu Gelb. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 2 nm gemessen. Der OD-Wert ist proportional zur Konzentration von Maus bFGF/FGF2. Sie können die Konzentration von Maus bFGF/FGF2 in den Proben berechnen, indem Sie die OD der Proben mit der Standardkurve vergleichen.
