Technical note
Dieses ELISA-Kit verwendet das kompetitive ELISA-Prinzip. Die in diesem Kit enthaltene Mikro-ELISA-Platte wurde mit humanem IgG2 vorbeschichtet. Während der Reaktion konkurriert humanes IgG2 in der Probe oder im Standard mit einer festen Menge an humanem IgG2 auf dem festphasengebundenen Träger um Bindungsstellen am biotinylierten Nachweis-Antikörper, der spezifisch für humanes IgG2 ist. Überschüssiges Konjugat und ungebundene Probe oder Standard werden abgewaschen, und Avidin-Meerrettichperoxidase (HRP)-Konjugat wird zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Anschließend wird eine TMB-Substratlösung zu jeder Vertiefung gegeben. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Stopplösung beendet, wobei sich die Farbe von blau nach gelb ändert. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 2 nm gemessen. Die Konzentration von humanem IgG2 in den getesteten Proben kann durch Vergleich der OD der Proben mit der Standardkurve berechnet werden.
