Technical note
Dieses ELISA-Kit basiert auf dem Prinzip des kompetitiven ELISA. Die in diesem Kit enthaltene Mikro-ELISA-Platte ist mit humanem αMSH vorbeschichtet. Während der Reaktion konkurriert humanes αMSH in der Probe oder dem Standard mit einer festgelegten Menge an humanem αMSH auf dem Festphasenträger um Bindungsstellen am biotinylierten Detektionsantikörper, der spezifisch für humanes αMSH ist. Überschüssiges Konjugat und ungebundene Proben oder Standards werden abgewaschen, und Avidin-Meerrettichperoxidase (HRP)-Konjugat wird zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Anschließend wird eine TMB-Substratlösung zu jeder Vertiefung gegeben. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Stopplösung beendet, und die Farbe wechselt von blau zu gelb. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 2 nm gemessen. Die Konzentration des humanen αMSH in den getesteten Proben kann durch Vergleich der OD der Proben mit der Standardkurve berechnet werden.
