Technical note
Dieses ELISA-Kit basiert auf dem Prinzip des kompetitiven ELISA. Die in diesem Kit enthaltene Mikro-ELISA-Platte ist mit humanem GLP-2 vorbeschichtet. Während der Reaktion konkurriert das humane GLP-2 in der Probe oder dem Standard mit einer festen Menge an humanem GLP-2 auf dem Festphasenträger um die Bindungsstellen am biotinylierten Detektionsantikörper, der spezifisch für humanes GLP-2 ist. Überschüssiges Konjugat und ungebundene Probe oder Standard werden abgewaschen, und Avidin-Meerrettichperoxidase (HRP)-Konjugat wird zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Anschließend wird eine TMB-Substratlösung zu jeder Vertiefung gegeben. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Stopplösung beendet, und die Farbe wechselt von blau zu gelb. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 2 nm gemessen. Die Konzentration von humanem GLP-2 in den getesteten Proben kann durch Vergleich der OD der Proben mit der Standardkurve berechnet werden.
