Technical note
Dieser ELISA-Kit verwendet das kompetitive ELISA-Prinzip. Die in diesem Kit enthaltene Mikro-ELISA-Platte ist mit humanem TXB2 vorbeschichtet. Während der Reaktion konkurriert humanes TXB2 in der Probe oder dem Standard mit einer festen Menge an humanem TXB2 auf dem Festphasenträger um die Bindungsstellen am biotinylierten Detektionsantikörper, der spezifisch für humanes TXB2 ist. Überschüssiges Konjugat und ungebundene Proben oder Standards werden abgewaschen, und Avidin-Meerrettichperoxidase (HRP)-Konjugat wird zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Anschließend wird eine TMB-Substratlösung zu jeder Vertiefung hinzugefügt. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Stopplösung beendet, und die Farbe wechselt von blau zu gelb. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 2 nm gemessen. Die Konzentration von humanem TXB2 in den getesteten Proben kann durch Vergleich der OD der Proben mit der Standardkurve berechnet werden.
