Technical note
Dieses ELISA-Kit verwendet das Sandwich-ELISA-Prinzip. Die in diesem Kit enthaltene Mikro-ELISA-Platte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Human SEMA3A ist. Standards oder Proben werden zu den Mikro-ELISA-Plattenvertiefungen gegeben und mit dem spezifischen Antikörper kombiniert. Anschließend werden nacheinander ein biotinylierter Detektionsantikörper, der spezifisch für Human SEMA3A ist, und ein Avidin-Meerrettichperoxidase (HRP)-Konjugat zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Freie Komponenten werden abgewaschen. Die Substratlösung wird zu jeder Vertiefung gegeben. Nur die Vertiefungen, die Human SEMA3A, den biotinylierten Detektionsantikörper und das Avidin-HRP-Konjugat enthalten, werden blau gefärbt. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Stopplösung beendet und die Farbe wird gelb. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 2 nm gemessen. Der OD-Wert ist proportional zur Konzentration von Human SEMA3A. Sie können die Konzentration von Human SEMA3A in den Proben berechnen, indem Sie die OD der Proben mit der Standardkurve vergleichen.
