Technical note
Dieses ELISA-Kit verwendet das Sandwich-ELISA-Prinzip. Die in diesem Kit enthaltene Mikro-ELISA-Platte ist mit einem Antikörper vorbeschichtet, der spezifisch für menschliches COL1α2 ist. Standards oder Proben werden in die Vertiefungen der Mikro-ELISA-Platte gegeben und mit dem spezifischen Antikörper verbunden. Anschließend werden nacheinander ein biotinylierter Detektionsantikörper, der spezifisch für menschliches COL1α2 ist, und ein Avidin-Meerrettichperoxidase (HRP)-Konjugat zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Freie Komponenten werden abgewaschen. Die Substratlösung wird zu jeder Vertiefung gegeben. Nur jene Vertiefungen, die menschliches COL1α2, einen biotinylierten Detektionsantikörper und ein Avidin-HRP-Konjugat enthalten, erscheinen blau. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Stopplösung beendet, und die Farbe wechselt zu gelb. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 2 nm gemessen. Der OD-Wert ist proportional zur Konzentration von menschlichem COL1α2. Die Konzentration von menschlichem COL1α2 in den Proben kann durch Vergleich der OD der Proben mit der Standardkurve berechnet werden.
