Technical note
Dieses ELISA-Kit nutzt das Sandwich-ELISA-Prinzip. Die in diesem Kit enthaltene Mikro-ELISA-Platte ist mit einem Antikörper vorbeschichtet, der spezifisch für humanes BMPR2 ist. Standards oder Proben werden in die Vertiefungen der Mikro-ELISA-Platte gegeben und mit dem spezifischen Antikörper kombiniert. Anschließend werden nacheinander ein biotinylierter Detektionsantikörper, der spezifisch für humanes BMPR2 ist, und ein Avidin-Meerrettichperoxidase (HRP)-Konjugat zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Freie Komponenten werden abgewaschen. Die Substratlösung wird zu jeder Vertiefung gegeben. Nur jene Vertiefungen, die humanes BMPR2, biotinylierten Detektionsantikörper und Avidin-HRP-Konjugat enthalten, erscheinen blau. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Stopplösung beendet und die Farbe wechselt zu Gelb. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 2 nm gemessen. Der OD-Wert ist proportional zur Konzentration von humanem BMPR2. Sie können die Konzentration von humanem BMPR2 in den Proben berechnen, indem Sie die OD der Proben mit der Standardkurve vergleichen.

BMPR2 (bone morphogenetic protein receptor type 2; BMPR-II/BMPR-2) is a type II transmembrane serine/threonine kinase receptor in BMP signalling. In ligand-induced receptor complexes, BMPR2 activates type I BMP receptors, enabling downstream SMAD transcriptional responses.

Biological function: BMPR2 is a type II receptor serine/threonine kinase that binds BMP-family ligands including BMP2 and BMP7. Upon ligand-dependent receptor-complex formation, BMPR2 phosphorylates and activates type I BMP receptors; activated type I receptors then engage SMAD transcriptional regulators. Curated UniProt evidence also supports non-canonical signalling through the p38 MAPK cascade in defined contexts.
