Technical note
Dieses ELISA-Kit basiert auf dem Sandwich-ELISA-Prinzip. Die in diesem Kit enthaltene Mikro-ELISA-Platte wurde mit einem Antikörper vorbeschichtet, der spezifisch für humanes ADAM10 ist. Standards oder Proben werden in die Vertiefungen der Mikro-ELISA-Platte gegeben und mit dem spezifischen Antikörper kombiniert. Anschließend werden nacheinander ein biotinylierter Nachweisantikörper, der spezifisch für humanes ADAM10 ist, und ein Avidin-Meerrettichperoxidase (HRP)-Konjugat zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nicht gebundene Komponenten werden abgewaschen. Die Substratlösung wird zu jeder Vertiefung gegeben. Nur die Vertiefungen, die humanes ADAM10, biotinylierten Nachweisantikörper und Avidin-HRP-Konjugat enthalten, färben sich blau. Die Enzym-Substrat-Reaktion wird durch Zugabe der Stopplösung beendet, woraufhin sich die Farbe gelb färbt. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 2 nm gemessen. Der OD-Wert ist proportional zur Konzentration von humanem ADAM10. Die Konzentration von humanem ADAM10 in den Proben kann durch Vergleich der OD der Proben mit der Standardkurve berechnet werden.
