Technical note
Dieses ELISA-Kit nutzt das kompetitive ELISA-Prinzip. Die in diesem Kit enthaltene Mikro-ELISA-Platte wurde mit humanem ACTH vorbeschichtet. Während der Reaktion konkurriert humanes ACTH in der Probe oder im Standard mit einer festen Menge an humanem ACTH auf dem Festphasenträger um Bindungsstellen am biotinylierten Detektionsantikörper, der spezifisch für humanes ACTH ist. Überschüssiges Konjugat und ungebundene Proben oder Standards werden abgewaschen, und Avidin-Meerrettichperoxidase (HRP)-Konjugat wird zu jeder Mikrotiterplatte hinzugefügt und inkubiert. Anschließend wird eine TMB-Substratlösung zu jeder Vertiefung gegeben. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Stopplösung beendet, und die Farbe wechselt von blau zu gelb. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 2 nm gemessen. Die Konzentration des humanen ACTH in den getesteten Proben kann durch Vergleich der OD der Proben mit der Standardkurve berechnet werden.
