Dieser ELISA-Kit verwendet Competitive-ELISA als Methode zum qualitativen Nachweis des Anti-SARS-CoV-2-Neutralisationsantikörpers in der Probe. _x005F_x000D_ Die in diesem Kit enthaltene Mikro-ELISA-Platte ist mit rekombinantem humanem ACE2 vorbeschichtet. Während der Reaktion konkurriert der SARS-CoV-2-Neutralisationsantikörper in den vorbehandelten Proben oder Kontrollen mit einer festgelegten Menge an humanem ACE2 auf dem festen Träger um Bindungsstellen am Meerrettichperoxidase (HRP)-konjugierten rekombinanten SARS-CoV-2-RBD-Fragment (HRP-RBD). Nach 37°C Inkubation werden das ungebundene HRP-RBD sowie jedes HRP-RBD, das an Nicht-Neutralisationsantikörper gebunden ist, auf der Platte eingefangen und bilden schließlich den ACE2-RBD-HRP-Komplex, während die zirkulierenden Neutralisationsantikörper-HRP-RBD-Komplexe im Überstand verbleiben und während des Waschvorgangs entfernt werden. Anschließend wird jedem Well eine TMB-Substratlösung hinzugefügt. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Stopplösung beendet und die Farbänderung spektrophotometrisch bei einer Wellenlänge von 450 nm ± 2 nm gemessen. Anhand des Hemmverhältnisses wird beurteilt, ob SARS-CoV-2-Neutralisationsantikörper in den getesteten Proben vorhanden sind oder nicht.
