Technical note
Dieser ELISA-Kit verwendet das Sandwich-Ag-ELISA-Prinzip. Die in diesem Kit enthaltene Mikro-ELISA-Platte wurde mit rekombinantem SARS-CoV-2-Spike-Protein vorbeschichtet. Nach Zugabe von Proben (oder Kontrollen) zu den Vertiefungen verbinden sich die Gesamtantikörper gegen SARS-CoV-2-Spike-Protein mit dem vorbeschichteten SARS-CoV-2-Spike-Protein. Nach vollständigem Waschen wird Meerrettichperoxidase (HRP)-konjugiertes SARS-CoV-2-Spike-Protein hinzugefügt, um den Antigen-Antikörper-HRP-konjugierten Antigenkomplex zu entwickeln. Freie Komponenten werden abgewaschen. Die Substratlösung wird jeder Vertiefung hinzugefügt. Nur jene Vertiefungen, die SARS-CoV-2-Spike-Protein, Antikörper gegen SARS-CoV-2-Spike-Protein und HRP-konjugiertes SARS-CoV-2-Spike-Protein enthalten, erscheinen blau. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Stopplösung beendet, woraufhin sich die Farbe gelb färbt. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450 ± 2 nm gemessen. Dies wird mit dem CUT OFF-Wert verglichen, um festzustellen, ob SARS-CoV-2-Spike-Protein-Gesamtantikörper in den getesteten Proben vorhanden sind oder nicht.
