Technical note
Dieser ELISA-Kit verwendet das indirekte ELISA-Verfahren zum qualitativen Nachweis von SARS-CoV-2 Nukleokapsid-Protein-IgM in der Probe. Die in diesem Kit enthaltene Mikro-ELISA-Platte ist mit gereinigtem SARS-CoV-2 Nukleokapsid-Protein-Antigen (E-ELC-001) vorbeschichtet. Nach Zugabe der Proben zu den Wells bindet das SARS-CoV-2 Nukleokapsid-Protein-IgM in den Proben an das vorbeschichtete SARS-CoV-2 Nukleokapsid-Protein-Antigen. Nach vollständigem Waschen wird Meerrettichperoxidase (HRP)-konjugiertes Maus-Anti-Human-IgM hinzugefügt, um den Antigen-Antikörper-HRP-konjugierten Sekundärantikörperkomplex zu entwickeln. Freie Komponenten werden abgewaschen, dann wird die Substratlösung zu jedem Well gegeben. Nur Wells, die SARS-CoV-2 Nukleokapsid-Protein-IgM und HRP-konjugiertes Anti-Human-IgM enthalten, erscheinen blau. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Stopplösung beendet und die Farbe wird gelb. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450 ± 2 nm gemessen. Durch den Vergleich mit dem CUT OFF-Wert wird beurteilt, ob SARS-CoV-2 Nukleokapsid-Protein-IgM in den getesteten Proben vorhanden ist oder nicht.
