Technical note
Dieses ELISA-Kit basiert auf dem Prinzip des kompetitiven ELISA. Die in diesem Kit enthaltene Mikro-ELISA-Platte ist mit Universal-PGI2 vorbeschichtet. Während der Reaktion konkurriert Universal-PGI2 in der Probe oder dem Standard mit einer fixierten Menge an Universal-PGI2 auf dem Festphasenträger um die Bindungsstellen am biotinylierten Nachweisantikörper, der spezifisch für Universal-PGI2 ist. Überschüssiges Konjugat und ungebundene Probe oder Standard werden abgewaschen, und Avidin-Meerrettichperoxidase (HRP)-Konjugat wird zu jeder Mikrotiterplatte hinzugefügt und inkubiert. Anschließend wird eine TMB-Substratlösung zu jeder Vertiefung gegeben. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Stopplösung beendet, wobei sich die Farbe von blau nach gelb ändert. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 2 nm gemessen. Die Konzentration von Universal-PGI2 in den getesteten Proben kann durch Vergleich der OD der Proben mit der Standardkurve berechnet werden.
