Technical note
Dieser CLIA-Kit verwendet das Sandwich-CLIA-Prinzip. Die in diesem Kit enthaltene Mikro-CLIA-Platte wurde mit einem Antikörper vorbeschichtet, der spezifisch für humanes PLAU/uPA ist. Proben oder Standards werden in die Vertiefungen der Mikro-CLIA-Platte gegeben und mit dem spezifischen Antikörper kombiniert. Anschließend werden nacheinander ein biotinylierter Nachweisantikörper, der spezifisch für humanes PLAU/uPA ist, und ein Avidin-Meerrettichperoxidase (HRP)-Konjugat zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Freie Komponenten werden abgewaschen. Die Substratlösung wird zu jeder Vertiefung gegeben. Nur die Vertiefungen, die humanes PLAU/uPA, den biotinylierten Nachweisantikörper und das Avidin-HRP-Konjugat enthalten, zeigen Lumineszenz. Der Wert der relativen Lichteinheit (RLU) wird mit einem Chemilumineszenz-Immunoassay-Analysator bei 425 nm gemessen. Der RLU-Wert ist positiv mit der Konzentration von humanem PLAU/uPA assoziiert. Die Konzentration von humanem PLAU/uPA in den Proben kann durch Vergleich des RLU-Wertes der Proben mit der Standardkurve berechnet werden.
