Technical note
Dieses CLIA-Kit verwendet das Sandwich-CLIA-Prinzip. Die in diesem Kit enthaltene Mikro-CLIA-Platte wurde mit einem für humanes SEMA3A spezifischen Antikörper vorbeschichtet. Proben oder Standards werden zu den Vertiefungen der Mikro-CLIA-Platte gegeben und mit dem spezifischen Antikörper kombiniert. Anschließend werden nacheinander ein biotinylierter Detektionsantikörper, der spezifisch für humanes SEMA3A ist, und Avidin-Meerrettichperoxidase (HRP)-Konjugat zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Freie Komponenten werden abgewaschen. Die Substratlösung wird zu jeder Vertiefung gegeben. Nur jene Vertiefungen, die humanes SEMA3A, biotinylierten Detektionsantikörper und Avidin-HRP-Konjugat enthalten, zeigen Lumineszenz. Der Wert der relativen Lichteinheit (RLU) wird mit einem Chemilumineszenz-Immunoassay-Analysator bei 425 nm gemessen. Der RLU-Wert ist positiv mit der Konzentration von humanem SEMA3A assoziiert. Die Konzentration von humanem SEMA3A in den Proben kann durch Vergleich des RLU-Werts der Proben mit der Standardkurve berechnet werden.
