Technical note
Dieser CLIA-Kit nutzt das Sandwich-CLIA-Prinzip. Die in diesem Kit enthaltene Mikro-CLIA-Platte ist mit einem Antikörper vorbeschichtet, der spezifisch für humanes MYO ist. Proben oder Standards werden in die Vertiefungen der Mikro-CLIA-Platte gegeben und mit dem spezifischen Antikörper kombiniert. Anschließend werden nacheinander ein biotinylierter Detektionsantikörper, der spezifisch für humanes MYO ist, und ein Avidin-Meerrettichperoxidase (HRP)-Konjugat zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nicht gebundene Komponenten werden abgewaschen. Die Substratlösung wird zu jeder Vertiefung gegeben. Nur jene Vertiefungen, die humanes MYO, den biotinylierten Detektionsantikörper und das Avidin-HRP-Konjugat enthalten, zeigen Lumineszenz. Der Wert der relativen Lichteinheit (RLU) wird vom Chemilumineszenz-Immunoassay-Analysator bei 425 nm gemessen. Der RLU-Wert ist positiv mit der Konzentration von humanem MYO assoziiert. Sie können die Konzentration von humanem MYO in den Proben berechnen, indem Sie den RLU-Wert der Proben mit der Standardkurve vergleichen.
