Technical note
Annexin V gehört zur Annexin-Familie und bindet Phosphatidylserin (PS) kalziumabhängig. Das mit Fluoreszein markierte Annexin V bindet spezifisch an das PS auf der äußeren Membranseite von apoptotischen Zellen und kann mittels Durchflusszytometrie oder Fluoreszenzmikroskopie nachgewiesen werden. Kernfarbstoffe (PI, DAPI, 7-AAD) haben eine hohe DNA-Bindungskonstante und werden von intakten Zellen effizient ausgeschlossen. Sie sind nützlich für die DNA-Analyse und die Unterscheidung toter Zellen während der durchflusszytometrischen Analyse. Aufgrund später Apoptose oder des Verlusts der Membranintegrität in nekrotischen Zellen können Kernfarbstoffe in die Zellen eindringen, um die DNA zu färben. In Kombination mit Annexin V können Zellen in verschiedenen apoptotischen Stadien unterschieden werden.
