L-Äpfelsäure, auch bekannt als 2-Hydroxysuccinylsäure, ist in lebenden Organismen weit verbreitet. Sie hat zwei Stereoisomere und ist eines der Mitglieder des Tricarbonsäurezyklus, einem wichtigen Stoffwechselzyklus in lebenden Organismen. Das Nachweisprinzip dieses Kits besteht darin, dass das Enzym Äpfelsäure zu Oxalessigsäure katalysiert. Gleichzeitig wird unter Einwirkung eines elektronischen Kopplers das chromogene Reagenz zu einem orange-gelben Produkt reduziert, das bei etwa 450 nm eine maximale Absorptionsspitze aufweist.
