Triglyceride (TG) können durch Lipoproteinlipase in Glycerin und freie Fettsäuren hydrolysiert werden. Glycerin bildet unter der Katalyse von Glycerinkinase (GK) Glycerin-3-phosphat und ADP. Glycerin-3-phosphat erzeugt unter der Wirkung von Glycerinphosphatoxidase (GPO) Wasserstoffperoxid. In Gegenwart von Trinder-Reagenz wird Wasserstoffperoxid durch Peroxidase katalysiert, um Chinone zu bilden, deren Menge proportional zum TG-Gehalt ist.
