Die Probe wird hydrolysiert, um freies HYP zu erzeugen, und Hydroxyprolin kann unter der Einwirkung eines Oxidationsmittels ein Oxidationsprodukt bilden. Das entstandene Oxidationsprodukt kann mit einem Farbbildner reagieren und eine burgunderrote Farbe erzeugen. Die Konzentration von Hydroxyprolin kann durch Messung des OD-Wertes bei 558 nm berechnet werden.
