Monoaminoxidase (MAO) ist in den Mitochondrien von Leber-, Nieren-, Gehirn-, Dünndarm- und anderen Organzellen weit verbreitet und hauptsächlich an der metabolischen Inaktivierung bioaktiver Aminosubstanzen beteiligt. MAO wird aufgrund der Bindungsspezifität von Substraten oder Inhibitoren, der Zellverteilung und der Immunspezifität in zwei Subtypen unterteilt: MAO-A und MAO-B. MAO kann das entsprechende Substrat katalysieren, um H2O2 zu erzeugen, und H2O2 bildet unter Einwirkung des Enzyms einen roten Komplex mit der Fluoreszenzsonde. Die Änderung des Fluoreszenzwertes der Probe bei einer Anregungswellenlänge von 535 nm und einer Emissionswellenlänge von 587 nm pro Zeiteinheit kann gemessen werden, und die Aktivität von MAO-A und MAO-B der Probe kann durch Hemmung mit spezifischen Inhibitoren nachgewiesen und berechnet werden.
