Technical note
Dieser ELISA-Kit nutzt das Sandwich-ELISA-Prinzip. Die in diesem Kit enthaltene Mikro-ELISA-Platte ist mit einem Antikörper vorbeschichtet, der spezifisch für humanes IL-2 ist. Proben (oder Standards & Qualitätskontrollmaterial) werden in die Vertiefungen der Mikro-ELISA-Platte gegeben und mit dem spezifischen Antikörper kombiniert. Anschließend werden nacheinander ein biotinylierter Detektionsantikörper, der spezifisch für humanes IL-2 ist, und ein Avidin-Meerrettichperoxidase (HRP)-Konjugat zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Freie Komponenten werden abgewaschen. Die Substratlösung wird zu jeder Vertiefung gegeben. Nur jene Vertiefungen, die humanes IL-2, biotinylierten Detektionsantikörper und Avidin-HRP-Konjugat enthalten, werden blau gefärbt erscheinen. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Stopplösung beendet und die Farbe wechselt zu gelb. Die optische Dichte (OD) wird spektrophotometrisch bei einer Wellenlänge von 450 ± 3 nm gemessen. Der OD-Wert ist proportional zur Konzentration des humanen IL-2. Die Konzentration des humanen IL-2 in den Proben kann durch Vergleich der OD der Proben mit der Standardkurve berechnet werden.
