Test principle

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Human BCL2L11. Standards or samples are added to the appropriate microtiter plate wells then with a biotin-conjugated antibody specific to Human BCL2L11. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Human BCL2L11, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Human BCL2L11 in the samples is then determined by comparing the OD of the samples to the standard curve.

BCL2L11 (BCL2 like 11), commonly called BIM or Bcl-2-like protein 11, is a BH3-only BCL-2-family regulator that helps initiate mitochondrial apoptosis in response to developmental and cellular stress signals.

Biological function: BCL2L11 encodes a BH3-only pro-apoptotic BCL-2-family protein. BIM engages anti-apoptotic BCL-2-family members and helps shift mitochondrial signalling toward activation of the intrinsic apoptosis machinery. Alternative splicing generates multiple BCL2L11 isoforms whose abundance and activity are regulated by cell state and signalling inputs.
