SMAD3 is a receptor-regulated SMAD transcriptional mediator in the TGF-beta and activin signalling pathways. Following receptor-dependent activation, SMAD3 forms complexes including SMAD4 and contributes to transcriptional control of genes involved in processes such as cell proliferation, differentiation and tissue responses. SMAD3 is found in the cytoplasm and nucleus, with TGF-beta signalling promoting its accumulation in the nucleus as part of active SMAD complexes.
