The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Human AIF1. Standards or samples are added to the appropriate microtiter plate wells then with a biotin-conjugated antibody specific to Human AIF1. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Human AIF1, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Human AIF1 in the samples is then determined by comparing the OD of the samples to the standard curve.

AIF1 is allograft inflammatory factor 1, widely recognised in research as IBA1 (ionized calcium-binding adapter molecule 1). It is a calcium- and actin-binding protein associated with myeloid-cell cytoskeletal responses and is commonly studied in macrophage and microglial biology.
Biological function: AIF1/IBA1 binds actin and calcium and contributes to cytoskeletal remodelling, membrane ruffling and cell motility. Reviewed evidence supports roles in RAC-dependent actin dynamics, phagocytic activity and macrophage activation, making AIF1 useful for investigating changes in myeloid-cell state and morphology.
