Human C3(Complement Component 3) ELISA Kit is a research-use ELISA kit from ELK Biotechnology for measuring C3 in Serum, Plasma, Other biological fluids samples from Human. Use the listed assay range, sample guidance and protocol documents to assess suitability for the intended study.

Test principle

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Human C3. Standards or samples are added to the appropriate microtiter plate wells then with a biotin-conjugated antibody specific to Human C3. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Human C3, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Human C3 in the samples is then determined by comparing the OD of the samples to the standard curve.

C3 (complement C3) is the central complement precursor cleaved into C3a and C3b, linking complement activation to inflammatory signalling, opsonization and downstream C5 activation.

Biological function: Complement C3 is activated by C3 convertases across the classical, lectin and alternative pathways. Cleavage generates C3a, an anaphylatoxin involved in inflammatory signalling, and C3b, which covalently labels surfaces for opsonization, supports complement amplification and contributes to C5 convertase formation. Additional processing generates iC3b and other fragments with distinct immune functions.
