APEX1 (apurinic/apyrimidinic endodeoxyribonuclease 1; APE1/Ref-1) is the major human AP endonuclease and a multifunctional DNA-repair and redox regulator. It repairs abasic sites in base-excision repair and helps regulate redox-sensitive transcription-factor activity.
Biological function: APEX1 cleaves the DNA phosphodiester backbone immediately 5′ to apurinic/apyrimidinic sites generated spontaneously or during base-excision repair, creating DNA ends for downstream repair. Reviewed UniProt also supports a redox-regulatory role toward transcription factors, linking genome maintenance with oxidative-stress signalling.
