The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Human ACO1. Standards or samples are added to the appropriate microtiter plate wells then with a biotin-conjugated antibody specific to Human ACO1. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Human ACO1, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Human ACO1 in the samples is then determined by comparing the OD of the samples to the standard curve.

Aconitase 1 (ACO1), also known as cytoplasmic aconitate hydratase and iron regulatory protein 1 (IRP1), is a bifunctional human protein linking citrate metabolism with iron-responsive post-transcriptional regulation.
Biological function: ACO1 is a bifunctional cytosolic protein. When assembled with a [4Fe-4S] cluster it acts as aconitase, catalysing the reversible interconversion of citrate and isocitrate through cis-aconitate. Under iron-limiting conditions, loss of the cluster enables ACO1/IRP1 to bind iron-responsive elements in target mRNAs and regulate proteins involved in cellular iron uptake, storage and use.
