Use the listed assay range, sample guidance and protocol documents to assess suitability for the intended study.

Test principle

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Cattle CA1. Standards or samples are added to the appropriate microtiter plate wells then with a biotin-conjugated antibody specific to Cattle CA1. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Cattle CA1, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Cattle CA1 in the samples is then determined by comparing the OD of the samples to the standard curve.

Carbonic anhydrase 1 (CA1; carbonic anhydrase I/CA-I, also CAB) is a cytosolic zinc metalloenzyme that catalyses reversible carbon dioxide hydration. It is especially abundant in erythrocytes and is studied in CO2 transport, bicarbonate chemistry and systemic acid-base physiology.

Biological function: CA1 catalyses the reversible interconversion of carbon dioxide and water with bicarbonate and protons through a zinc-containing active site. This rapid reaction supports carbon dioxide handling and proton/bicarbonate balance; its high erythrocyte abundance makes CA1 useful for studying carbonic-anhydrase catalysis and red-cell gas and acid-base chemistry.
