Product description

Rat ACSL4(Acyl-CoA Synthetase Long Chain Family Member 4) ELISA Kit is a research-use ELISA kit from ELK Biotechnology for measuring ACSL4 in Serum, Plasma, Tissue homogenates, Cell lysates, Cell culture supernatant, Other biological fluids samples from Rat. Use the listed assay range, sample guidance and protocol documents to assess suitability for the intended study.

Test principle

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Rat ACSL4. Standards or samples are added to the appropriate microtiter plate wells then with a biotin-conjugated antibody specific to Rat ACSL4. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Rat ACSL4, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Rat ACSL4 in the samples is then determined by comparing the OD of the samples to the standard curve.

Research target context

ACSL4acyl-CoA synthetase long chain family member 4

View target guide

Acyl-CoA synthetase long chain family member 4 (ACSL4), also known as FACL4 and long-chain-fatty-acid--CoA ligase 4, activates long-chain fatty acids to acyl-CoA esters. Human ACSL4 maps to NCBI Gene 2182 and reviewed UniProtKB O60488; this record links the verified target to related ABMIUM catalogue products.

Biological function: ACSL4 catalyses activation of long-chain fatty acids to fatty acyl-CoA esters for cellular lipid synthesis and fatty-acid degradation. NCBI and reviewed UniProt describe a preference for arachidonate; UniProt also identifies ACSL4 as a ferroptosis activator by generating activated polyunsaturated-fatty-acid substrates that can undergo lipid peroxidation.
