Biological function

ATG5 is covalently conjugated to ATG12 through an autophagy-specific ubiquitin-like conjugation pathway. The ATG12-ATG5 complex, together with ATG16L1, provides ATG8-family ligase activity that promotes LC3/GABARAP lipidation on forming autophagosomal membranes and is required for efficient autophagosome assembly.

ATG5 is a core target in macroautophagy and autophagosome-formation studies, including experiments on ATG8/LC3 lipidation, autophagic flux and mitochondrial quality control. ATG5 loss or perturbation is commonly used to distinguish autophagy-dependent phenotypes, but interpretation should account for cell type and non-canonical ATG-protein functions.
