ATG4B is a peptidase family C54 cysteine protease that processes ATG8-family proteins, including LC3 and GABARAP family members, to expose the C-terminal glycine required for lipidation. It also removes lipid modifications from ATG8-family proteins, regulating autophagosome membrane dynamics and autophagic flux.
ATG4B is widely studied in macroautophagy because ATG8-family processing and delipidation influence LC3/GABARAP membrane association, autophagosome maturation and flux. Protease-activity, lipidation and autophagic-flux assays should be interpreted in the context of substrate specificity and other ATG4-family enzymes.
