ALOX5 catalyses conversion of arachidonic acid to 5-hydroperoxyeicosatetraenoic acid (5-HPETE) and then leukotriene A4 (LTA4), a branch-point intermediate for downstream leukotrienes. This places 5-LOX at a central enzymatic step in leukotriene generation.
ALOX5/5-LOX is widely investigated in leukotriene biosynthesis, inflammatory and allergic signalling, innate-immune lipid metabolism and pharmacological modulation of the 5-lipoxygenase pathway. Experimental interpretation should distinguish ALOX5 abundance, localisation and catalytic activity.
