BCR combines opposing regulatory activities toward small GTP-binding proteins. Its C-terminal GAP domain promotes GTP hydrolysis by RAC1, RAC2 and CDC42, while its central DH domain has guanine-nucleotide exchange activity toward CDC42, RHOA and RAC1. The amino terminus also has intrinsic kinase activity. BCR is additionally important because chromosomal rearrangement with ABL1 produces the BCR-ABL fusion associated with chronic myeloid leukaemia and other leukaemias.
BCR is studied in small-GTPase regulation, cytoskeletal signalling, cell motility and differentiation, and in the biology of BCR-ABL fusion-driven leukaemia. For reagent selection, distinguish assays intended for native BCR from those designed to detect a BCR-ABL fusion or a specific fusion breakpoint, and confirm species, epitope, application and product-level validation.
