BACE1 is a type I transmembrane aspartyl protease. It cleaves APP at the beta-secretase site, generating the C99 fragment that can be further processed by gamma-secretase to produce amyloid-beta peptides. BACE1 activity is favoured in acidic intracellular compartments such as endosomes, and the enzyme also processes additional neuronal membrane proteins.
BACE1 is investigated in proteolytic regulation of APP, amyloidogenic processing, endosomal trafficking and neuronal membrane-protein turnover. Experimental studies commonly examine BACE1 abundance, maturation or localisation, protease activity and changes in APP-derived fragments when probing this pathway.
