Rekombinantes 2019-nCoV Guanin-N7_meth (N-6His) ist ein rekombinantes Protein von ELK Biotechnology für Forschungszwecke.
Rekombinante 2019-nCoV Guanin-N7-Methyltransferase wird von unserem E. coli-Expressionssystem hergestellt und das Zielgen, das Ala1-Gln527 kodiert, wird mit einem 6His-Tag am N-Terminus exprimiert.
Das nichtstrukturelle Protein (nsp) 14 von SARS-CoV 2 wurde als Cap (Guanin-N7)-Methyltransferase (N7-MTase) identifiziert. Nsp14 von Coronaviren hat zwei verschiedene Aktivitäten: eine Exoribonuklease-Aktivität, die auf sowohl ssRNA als auch dsRNA in 3' zu 5' Richtung wirkt, und eine N7-Guanin-Methyltransferase-Aktivität. Es könnte an der Korrekturlesefähigkeit während der viralen RNA-Replikation und -Transkription beteiligt sein. GTP, dGTP sowie Cap-Analoga GpppG, GpppA und m7GpppG konnten durch nsp14 methyliert werden.
