Die 6-Phosphogluconat-Dehydrogenase ist die zweite Dehydrogenase im Pentosephosphatweg. Ein Mangel dieses Enzyms ist im Allgemeinen asymptomatisch, und die Vererbung dieser Störung ist autosomal dominant. Hämolyse resultiert aus einer kombinierten Insuffizienz von 6-Phosphogluconat-Dehydrogenase und 6-Phosphogluconolactonase, was auf einen Synergismus der beiden Enzymopathien hindeutet. Für dieses Gen wurden mehrere Transkriptvarianten gefunden, die verschiedene Isoformen kodieren.
