Sialinsäure-Synthase ist ein Enzym, das beim Menschen durch das NANS-Gen kodiert wird. Dieses Gen kodiert ein Enzym, das in den Biosynthesewegen von Sialinsäuren fungiert. In vitro verwendet das kodierte Protein N-Acetylmannosamin-6-phosphat und Mannose-6-phosphat als Substrate, um phosphorylierte Formen von N-Acetylneuraminsäure (Neu5Ac) bzw. 2-Keto-3-Desoxy-D-glycero-D-galacto-nononsäure (KDN) zu erzeugen; es zeigt jedoch eine wesentlich höhere Aktivität gegenüber dem Neu5Ac-Phosphatprodukt. In Insektenzellen führt die Expression dieses Gens zur Produktion von Neu5Ac und KDN. Dieses Gen ist mit dem E. coli Sialinsäure-Synthase-Gen neuB verwandt und kann die Sialinsäure-Synthase-Aktivität in einem E. coli neuB-negativen Mutanten teilweise wiederherstellen.
