CA2 catalyses the reversible interconversion of carbon dioxide and water with bicarbonate and protons. Its high catalytic efficiency supports rapid CO2/bicarbonate equilibration and acid-base homeostasis, and CA2 can participate in membrane-associated bicarbonate-transport complexes despite being primarily cytosolic.

CA2 is widely studied in carbonic-anhydrase catalysis, pH regulation, bicarbonate transport, renal acid-base handling and skeletal physiology. Because several carbonic anhydrase isozymes share related chemistry, isoform-specific detection and inhibitor selectivity are important experimental considerations.
